What is CO2 Incubator?

The CO2 incubator is a cultivation instrument for cell, tissue, and bacterial culture. It is key equipment necessary to develop immunology, oncology, genetics, and bioengineering. It is a device that simulates the growth environment of cells or tissues in vivo by simulating the growth environment of cells/tissues in the incubator. The CO2 incubator requires a stable temperature (37°C), a stable CO2 level (5%), a constant pH value (pH: 7.2-7.4), and a high relative saturated humidity (95%) to culture cells/tissues in vitro.

Water Jacket & Air Jacket

When purchasing a carbon dioxide incubator, there are two types of heating structures to choose from: air jacket heating and water jacket heating.

Water Jacket: The water-jacketed incubator maintains a constant temperature through an independent hot water compartment surrounding the internal tank. The unique water jacket design enables the incubator to more reliably maintain the accuracy and stability of the temperature in the incubator for a long time in the event of a power failure. If your experimental environment is not stable (such as useful power limitation, or frequent power outages) and you need to maintain stable culture conditions for a long time, a water-jacketed carbon dioxide incubator is your best choice.

Air Jacket: The air jacket heating system directly heats the gas in the box through the heater, and can quickly restore the temperature stability in the box when the door is frequently opened and closed. Therefore, it has the characteristics of fast heating and rapid temperature recovery, which is especially beneficial for short-term cultivation and cultivation that requires frequent opening and closing of the door. In addition, the air-jacketed design is simpler than the water-jacketed design for users and is easier to maintain and clean.

Features of CO2 Incubator

Applications of CO2 Incubator

CO2 incubators have been widely used at present, mainly for the cultivation of biological cells, tissues, and bacteria in medical and biological experiments. The cultivation of biological cells is mainly carried out by simulating the temperature, humidity, and pH value of the organism. That is to say, the fungi with strict requirements on temperature, humidity, and pH value need to use a CO2 incubator.

How to Choose CO2 Incubator?

‌Choosing a suitable CO2 incubator requires considering the following key factors:

  1. ‌Temperature control‌:

‌Heating method‌: Common heating methods include direct heating, water jacket and air jacket. Direct heating heating elements are distributed on all sides of the box body, and fans are required to optimize temperature distribution; water jacket uses an independent water jacket layer to surround the internal box body to maintain a constant temperature, and has excellent thermal insulation performance; air jacket is heated by a heater installed in the cavity around the culture chamber, and the heat transfer is fast, which is suitable for frequent opening and closing of the box door.

‌Temperature uniformity and stability‌: The ideal temperature field uniformity is ±0.2℃, and stable temperature control is essential for normal cell growth.

‌Temperature range‌: The general temperature range is 5°C to 50°C, and mammalian cell culture is usually set to 37°C.

  1. ‌CO₂ concentration control‌:

‌Sensor type‌: infrared sensor (IR) and thermal conductivity sensor (TC). Infrared sensors have high precision, high stability and high accuracy, and are not affected by changes in temperature and humidity, but the price is relatively high; thermal conductivity sensors are greatly affected by the environment. ‌

‌Concentration control accuracy‌: High-precision CO₂ concentration control can ensure the stability of the pH value in the culture medium, and the general accuracy is ±0.1% or higher, which is more reliable. ‌

  1. Humidity control‌:

‌Humidification method‌: natural evaporation humidification and humidification disk humidification. Natural evaporation humidification increases humidity by natural evaporation of water in a heated environment; humidification disk humidification generates water vapor by heating the water in the humidification disk. ‌

‌Humidity range and stability‌: The appropriate humidity range is generally 90%-95%, and stable humidity control helps prevent the culture medium from drying up or being over-moistened. ‌

  1. ‌Contamination control‌:

‌Sterilization method‌: UV sterilization, 90℃ wet heat sterilization, and 140℃ and above dry heat sterilization. Dry heat sterilization is highly efficient and can completely kill microorganisms. ‌

‌Air filtration system‌: High-efficiency air filtration system can prevent microbial contamination and ensure experimental safety.
